Malegra DXT is a combination medication containing sildenafil citrate (100 mg) and duloxetine hydrochloride (30 mg), primarily used to treat erectile dysfunction and premature ejaculation. Both active ingredients undergo hepatic metabolism but through different enzymatic pathways. Malegra DXT is rapidly absorbed after oral administration, with peak plasma concentrations occurring within 30 to 120 minutes. It is extensively metabolized in the liver by cytochrome P450 enzymes, primarily CYP3A4 and CYP2C9. The resulting metabolites are excreted in the urine and feces. Sildenafil has a half-life of approximately 4 hours. Duloxetine is also absorbed well after oral administration, reaching peak plasma levels within 2 hours. It is metabolized in the liver via multiple cytochrome P450 enzymes, including CYP1A2 and CYP2D6. Duloxetine has a longer half-life, ranging from 8 to 17 hours, and is excreted in the urine and feces.
